Efficacy of density gradient centrifugation technique (DGC) in enhancing sperm cell DNA quality for assisted reproductive technique

Abstract Objective Density gradient centrifugation (DGC) is commonly used for sperm preparation before assisted reproductive technology (ART) procedures. This technique separates superior motile spermatozoa with normal morphology from the total population. However, there still controversy as to effects of this separation on cell DNA integrity which a determining element in process fertilization and embryonic development. The objective study was determine DGC assessed by novel association between two cytogenetic tests. Study designs Semen samples were collected 30 fertile donors 40 patients being candidates ART treatment. Each sample divided into parts: first portion subjected selection layers (45% 90%) second fraction rinsed phosphate-buffered saline solution centrifuged without density gradient. Abnormal chromatin structure evaluated dispersion (SCD) test denaturation an acridine orange (AO) monitored initially washed different centrifugation. Results significantly improved proportion progressive motility, morphology. Moreover, following centrifugation, denaturated decreased when compared whole semen ( p < 0.001). In addition, we found that isolated 90% layer possessed lower percentage decondensation those remaining 45% unprocessed Conclusions Using double tests, our shows processing useful selecting higher double-strand recommended be reproduction.
